چکیده

DNA barcoding—sequencing a standard region of the mitochondrial cytochrome c oxidase 1 gene (COI)— promises a rapid, accurate means of identifying animals to a species level. This study establishes that sequence variability in the barcode region permits discrimination of 98% of 201 Wsh species from the Canadian PaciWc. The average sequence variation within species was 0.25%, while the average distance separating species within genera was 3.75%. The latter value was considerably lower than values reported in other studies, reXecting the dominance of the Canadian fauna by members of the young and highly diverse genus Sebastes. Although most sebastids possessed distinctive COI sequences, four species did not. As a partial oVset to these cases, the barcode records indicated the presence of a new, broadly distributed species of Paraliparis and the possibility that Paraliparis pectoralis is actually a species pair. The present study shows that most Wsh species in PaciWc Canadian waters correspond to a single, tightly cohesive array of barcode sequences that are distinct from those of any other species, but also highlights some taxonomic issues that need further investigation.

Preying on commercial Wsheries and accumulating paralytic shellWsh toxins: a dietary analysis of invasive Dosidicus gigas (Cephalopoda Ommastrephidae) stranded in PaciWc Canada

In fall of 2009, several mass strandings of Humboldt squid (Dosidicus gigas) occurred on Vancouver Island (49°7 60N 125°54 0W). Morphological dissections coupled with DNA barcoding of stomach contents revealed Sardinops sagax (PaciWc sardine) and Clupea pallasii (PaciWc herring) as their primary prey. Plastic nurdles, Wshing line, bull kelp, eelgrass, and a guillemot feather were also discovere...

Habitat choice, recruitment and the response of coral reef Wshes to coral degradation

The global degradation of coral reefs is having profound eVects on the structure and species richness of associated reef Wsh assemblages. Historically, variation in the composition of Wsh communities has largely been attributed to factors aVecting settlement of reef Wsh larvae. However, the mechanisms that determine how Wsh settlers respond to diVerent stages of coral stress and the extent of c...

Appraisal of the entire mitochondrial genome for DNA barcoding in birds

DNA barcoding based on a standardized region of 648 base pairs of mitochondrial DNAsequences from Cytochrome C Oxidase 1 (COX1) is proposed for animal species identification.Recent studies suggested that DNA barcoding has been effective for identifying 94% of birdspecies. The proposed threshold of 10 times the average intraspecific variation could be used forthe identification and delimitation ...

Identification of Crocus sativus and its Adulterants from Chinese Markets by using DNA Barcoding Technique

Background: Saffron (Crocus sativus L.) is a common but very expensive herbal medicine. As an important traditional medicine, it has an outstanding effect in treating irregular and painful menstruation. Recently, the over-demand tendency of saffron results in an unusual phenomenon in the medicinal markets. Adulterants and saffron-like substitutes are intentionally mixed into medicinal markets a...

DNA Barcoding: a new tool with wide array of applications

DNA barcoding is a new term introduced in to scientific literatures by Hebert and coworkers almost a decade ago. The concept of barcoding alone is well-known to the public: a series of black bars printed on many commercial products (Universal Product Code), which are used to distinguish different products. Advances made in molecular biology and molecular techniques late 20th century e.g. sequen...
